SonoThera raised $125 million in an oversubscribed Series B to advance ultrasound-mediated genetic medicines into the clinic. The round was led by Vida Ventures, with participation from Otsuka Pharmaceutical, Otsuka Ventures and others, and will support lead programs in Duchenne muscular dystrophy and autosomal dominant polycystic kidney disease. The platform combines RIPPLE for ultrasound-mediated delivery and PORE for payload engineering, with the company positioning its approach for DNA/RNA therapeutics, gene editing, and redosing—aiming to address delivery, payload size, immune, and safety constraints seen in conventional gene therapy modalities. SonoThera expects to initiate its first DMD clinical trial in 2027, indicating a fast follow-through from financing to clinical timing while scaling platform capabilities across additional organ systems.
