Skyhawk Therapeutics reported that its once-daily RNA splicing candidate, SKY-0515, improved clinical symptoms in an extension analysis tied to its early-stage Huntington’s trial program. The company said patients taking SKY-0515 saw an average 1.59-point improvement on a combined functional/cognitive/movement scale versus an untreated comparison group from a natural history database. Skyhawk also reported target engagement signals, stating that at higher doses it lowered mutant huntingtin protein by more than 60% and reduced PMS1 messenger RNA by 25% throughout the study. The company said the therapy was generally well tolerated, with no serious adverse events reported. The extension readout strengthens the rationale for SKY-0515 as Skyhawk prepares larger studies, including one trial that has completed enrollment, as the field continues to watch for RNA-based approaches that combine symptom benefit with robust disease biology readouts.
