Silence Therapeutics reported positive Phase II results for divesiran, an siRNA targeting TMPRSS6 for polycythemia vera (PV). In the SANRECO trial (NCT05499013), the program met its primary endpoint, with an 88% response rate in the divesiran arms versus 19% in placebo-controlled groups. The company described hematocrit control as central to lowering thrombotic risk, and said divesiran reduced reliance on frequent phlebotomies. Silence reported that patients treated with divesiran went a total of 36 weeks without requiring phlebotomy, with dosing as infrequent as every three months. The update strengthens the case for a reversible, RNA-silencing mechanism that adjusts iron balance signals at the message level, aiming to reduce PV morbidity tied to blood-clot events.
