Researchers reported a human-protein approach to RNA base editing by engineering Argonaute 2 (AGO2) to deliver targeted C-to-U edits. In HEK293T cell experiments, the AGO2-based system achieved, on average, 1.9-fold higher C-to-U editing on mRNA targets than two Cas13 editor comparators. The study used guide RNAs designed to remodel target RNA structure and reduce nuclease activity, and then performed transcriptome-wide RNA-seq to estimate off-target editing. The authors reported similar or lower off-target editing compared with Cas13-based editors, alongside AI-assisted deimmunization considerations using predicted immunogenic peptide changes. The work adds a potential therapeutic engineering path that leverages endogenous human machinery rather than importing foreign scaffolds for programmable RNA manipulation.
