A first small phase I trial in severe malaria selected sevuparin 3 mg/kg as the maximum tolerated dose for further investigation. The study used continual reassessment based on coagulation readouts in hospitalized children, with dose-limiting coagulation abnormalities emerging at higher dose levels. In neuroinflammatory drug discovery, intranasal creatine prodrug CBT101 produced exploratory EEG changes consistent with central activity in a Phase 1 trial while remaining tolerated in healthy men. The readout extends prodrug-based CNS targeting beyond safety into functional biomarker signals. Together, the updates show early-stage translational efforts pushing for mechanistic confirmation—EEG for CNS activity and coagulation safety windows for adjunctive severe-malaria therapy.
