Silence Therapeutics reported positive Phase II results for divesiran, an siRNA targeting TMPRSS6 in polycythemia vera (PV). In the SANRECO trial (NCT05499013), the company said the program met its primary endpoint with an 88% response rate in treated patients versus 19% in placebo-controlled groups. Silencing TMPRSS6 is intended to reduce iron regulation and lower abnormal red blood cell production, with a clinical focus on maintaining hematocrit control while reducing reliance on frequent phlebotomies. Silence reported that treated patients spent a cumulative 36 weeks without requiring phlebotomy and that dosing intervals could extend to every three months. The results—if corroborated in further studies—could support a differentiation strategy for hematocrit management in PV, where thrombotic events remain a central morbidity and mortality driver.
