The transaction brings Alesta’s clinical-stage candidate ALE1—an orally active small molecule targeting inorganic pyrophosphate biology—into BioMarin’s pipeline. ALE1 is in an ongoing phase 1/2a study (NCT07179640) for hypophosphatasia, a rare genetic disorder caused by ALPL mutations that disrupt bone and tooth mineralization. BioMarin positioned the acquisition as a way to add an oral alternative to injectable therapies while expanding its addressable patient population. The acquisition also reflects how mineralization-focused programs remain strategically attractive in rare diseases, even as companies recalibrate portfolios following earlier pipeline decisions. The deal structure includes a spinout of non-ALE1 assets into a separate entity prior to closing.
