A multicenter single-arm Phase 2 trial in China reported an objective response rate of 70.5% at end of induction in previously untreated diffuse large B-cell lymphoma patients with high central nervous system relapse risk. The regimen combined orelabrutinib with R-CDOP across four centers. The study evaluated 44 patients and reported the 70.5% end-of-induction response rate as the headline efficacy outcome. The single-arm design means there is no randomized comparator to interpret relative benefit, but the dataset can still influence how oncology teams position BTK inhibition strategies in high-risk DLBCL. For pipeline tracking, the result adds to the growing set of BTK inhibitor-led combination approaches aimed at CNS-risk populations, where treatment intensification and regimen selection remain difficult. Confirmatory randomized studies would be needed to establish comparative effectiveness.
