Revolution Medicines reported Phase 3 results for daraxonrasib (RASolute 302) showing a substantial survival benefit in previously treated, metastatic pancreatic ductal adenocarcinoma. The trial, presented at ASCO 2026 and published concurrently in The New England Journal of Medicine, compared daraxonrasib against standard-of-care chemotherapy in 500 patients. Across the overall population, median survival was 13.2 months with daraxonrasib versus 6.7 months with chemotherapy. Disease progression was also delayed, with time to worsening of 7.2 months compared with 3.6 months for chemotherapy. The benefit was reported as broadly consistent in the subgroup with common RAS mutations as well, addressing a major clinical need in a cancer where outcomes remain poor and therapeutic options limited. The program targets the RAS pathway, a driver present in more than 90% of pancreatic tumors. The results raise the bar for subsequent-line pancreatic cancer development, setting up the next steps around regulatory strategy and positioning versus existing and emerging RAS-directed approaches.
