Rather than targeting central pain-signal processing like opioids, the program aims at a peripheral sensory-neuron pathway tied to the MNK-eIF4E signaling mechanism implicated in the shift from acute pain to chronic pain. 4E completed Phase I safety testing for 4E1103 (NCT07586514) and positioned the MNK approach as a way to pursue meaningful pain relief while reducing risks associated with central nervous system effects and addiction.
