Cretostimogene grenadenorepvec delivered strong complete response outcomes in a Phase 3 BOND-003 Cohort C study for high-risk, BCG-unresponsive non–muscle-invasive bladder cancer with carcinoma in situ. In the Lancet Oncology report, 75% of evaluable patients achieved a centrally confirmed complete response at any time after a median follow-up of 25.8 months. Responses continued to emerge after a second induction course for patients with persistent disease at 3 months, with 14 additional complete responses following re-induction. At 12 months, 46% remained in complete response, and 42% were still in complete response at 24 months. Safety results were predominantly mild and transient, supporting the continued development of the intravesical regimen delivered weekly for six weeks, with maintenance for responders. The study result provides an efficacy benchmark for communities seeking alternatives to BCG failure strategies and reinforces oncolytic immunotherapy as a potential next-line option for early-stage bladder cancer populations.
