A multi-method study in European Journal of Medical Research reported that TMCC2 suppresses osteosarcoma growth and links to M1 macrophage response biology. Across public TARGET-OS cohort analyses, higher TMCC2 expression associated with longer overall survival, while functional assays showed TMCC2 overexpression reduced viability, migration, and apoptosis-altering markers in U2OS and HOS osteosarcoma cell lines. Mechanistically, the study connected TMCC2-driven cellular effects to TGF-β1/SMAD2/3 signaling and found conditioned media from TMCC2-overexpressing cells increased CD86 and iNOS expression in THP-1-derived M1-like macrophages, suggesting a tumor–myeloid interaction axis.
