Tango Therapeutics reported updated Phase 1/2 results for vopimetostat, its PRMT5 inhibitor, combined with Revolution Medicines’ RAS(ON) asset daraxonrasib in metastatic, MTAP-deleted, RAS-mutant pancreatic ductal adenocarcinoma. At the August 20, 2026 data cutoff, Tango said the combination delivered an 82% objective response rate in 22 efficacy-evaluable patients with 2L/3L PDAC, with median progression-free survival not yet reached. The company also highlighted durability, citing a 9-month PFS rate of 88%, and said the regimen was generally well tolerated in the 30 PDAC and non-small cell lung cancer patients who received dose level 1. Tango presented the data at ESMO 2026 in Madrid and framed the results as supporting further development of the combination strategy. Tango said it plans to initiate a registrational Phase 3 study in 1H 2027 in front-line MTAP-deleted pancreatic cancer, with the design centered on the vopimetostat plus daraxonrasib backbone. CEO Malte Peters characterized the updated activity and tolerability as confirmation of an earlier June 2026 dataset, with the longer follow-up and broader population intended to strengthen the case for progression into late-stage testing. The read-through for biotech is that a chemotherapy-free targeted combination is gaining additional early clinical validation, putting pressure on the standard-of-care landscape where platinum-based or chemoimmunotherapy regimens have often remained central for advanced PDAC management.
