The iPRIME phase 2 trial evaluating neoadjuvant durvalumab plus tremelimumab alongside standard carboplatin and paclitaxel in advanced high-grade serous ovarian cancer did not meet its primary response endpoint. The study reported a 32.5% rate of chemotherapy response (CRS 3) in the immunotherapy arm, which fell short of the pre-specified threshold. Secondary readouts, including RECIST 1.1 radiologic response and progression-free survival, were broadly similar between the dual-immunotherapy regimen and chemotherapy alone. The result adds to the evidence base questioning the incremental value of upfront dual checkpoint blockade in this setting.
