Tango Therapeutics reported updated Phase 1/2 results for vopimetostat, a PRMT5 inhibitor, combined with Revolution Medicines’ RAS(ON) inhibitor daraxonrasib in advanced pancreatic ductal adenocarcinoma. In efficacy-evaluable patients with MTAP-deleted and RAS-mutant disease, the company reported an 82% objective response rate, with median progression-free survival not yet reached and a 9-month PFS rate of 88% as of the data cutoff. The program is being evaluated without chemotherapy, emphasizing targeted combinations aimed at exploiting tumor-specific biology. Management said the updated dataset confirmed an earlier June readout and supported progression discussions toward a registrational Phase 3 study in a defined front-line population. For the field, the update adds another proof point that epigenetic vulnerability (PRMT5 inhibition) and RAS pathway dependency may be combinable outside conventional backbone regimens, though larger randomized comparisons will be required to validate durability and survival impact.
