Roche has entered a global licensing and collaboration agreement with Zymeworks for ZW209, a trispecific T-cell engager targeting DLL3. Zymeworks will receive $55 million upfront, with an additional $40 million upon Phase Ib initiation, and eligibility for up to $718 million in further milestone payments plus tiered royalties. ZW209 is designed to engage T cells via CD3 and CD28 while targeting DLL3-expressing tumor cells, with conditional CD28 engagement and obligate cis-binding intended to reduce unintended T-cell activation. Zymeworks said the FDA has cleared the investigational new drug application for ZW209. For biotech and pharma strategists, the agreement is a signal of continuing expansion in multispecific cell-engager modalities, and it adds a new potential competitor to the DLL3-targeted landscape.
