Novo Nordisk agreed to an up to $1.4 billion multitarget partnership with Orbis Medicines, deepening its push toward oral biologics using synthetic macrocycles. Orbis’ macrocycle platform (“nCycles”) is designed to improve oral bioavailability and membrane permeability, targeting cardiometabolic diseases across validated targets. Under the agreement, Novo invests strategically in Orbis and could pay up-front and milestone payments plus tiered royalties tied to development and commercial achievements. Orbis’ pipeline initially focuses on targets validated by injectable blockbuster biologics. The deal reflects Novo’s effort to rejuvenate its cardiometabolic franchise after leadership losses in obesity and diabetes. It also underscores how “macrocycles as oral replacements for injectables” has become a key translational bet across peptide and peptide-mimetic technology. If successful, the partnership could produce next-generation oral candidates that address dose and delivery constraints seen in traditional oral peptide approaches.
