Circio and Tcelltech have agreed to collaborate on engineering next-generation in vivo CAR-T and TCR-T therapies using a non-integrating, non-viral vector strategy. The partners will combine Circio’s circVec expression technology with Tcelltech’s nanoSMAR DNA vector platform to deliver engineered gene expression without viral components. The staged program will start with proof-of-concept work comparing how durably the different technology combinations drive gene expression in primary human T cells. A subsequent functional phase will generate CD19-directed CAR T cells and test tumor-killing activity. The collaboration is positioned around safety and manufacturability advantages often associated with avoiding viral vectors, while leveraging nanoSMAR’s large cargo capacity to support complex constructs. Both companies framed the approach as aiming to improve sustained CAR/TCR expression and simplify manufacturing for wider accessibility.
