UCLA Health researchers reported that a single dose of rapamycin improved multiple autism-like features in a maternal inflammation mouse model, with effects appearing rapid but temporary. The team’s preclinical work, published in Nature Communications, targeted dysfunction linked to the mTOR pathway. Across measured behavioral and neural readouts—such as brain overactivity, seizure susceptibility, sensory sensitivity, repetitive behaviors, and abnormal functional network organization—rapamycin produced normalization within a short window in adulthood. The study also highlighted limitations for translation: repeated dosing lost efficacy and raised toxicity concerns. While rapamycin itself is not framed as a direct human candidate, the findings reinforce the idea that the adult brain may still be capable of functional normalization for certain autism-associated pathways.
