New research mapped how oral squamous cell carcinoma resists cisplatin by blocking ferroptosis. The secreted protein DKK1 was implicated in maintaining SLC3A2 cystine transporter activity through a beta-catenin/STAT3 pathway, suppressing ferroptotic cell death. In cell and mouse models, inhibiting DKK1 restored cisplatin sensitivity, positioning the axis as a potential combination strategy. The work also adds mechanistic clarity to ferroptosis modulation as a lever for overcoming platinum resistance. For oncology drug developers, the take-home is actionable: target DKK1-mediated transporter regulation to re-enable ferroptosis under chemotherapy pressure.
