Eli Lilly agreed to buy 4E Therapeutics, aiming to expand its pain pipeline with a non-opioid strategy that targets peripheral sensory neurons rather than centrally dulling pain signals. The Austin-based company’s approach centers on inhibiting the MNK-eIF4E signaling pathway, with 4ET1103 as its lead candidate. The acquisition follows Lilly’s earlier moves in the pain space, including its May 2025 purchase of SiteOne, and comes as developers continue to search for alternatives to opioid-based treatments. Lilly highlighted clinical development, translational, and global commercialization capabilities as a fit for advancing 4E’s program. For 4E, the deal provides resources to advance an oral MNK inhibitor that completed a Phase I safety study (NCT07586514), while Lilly’s broader M&A streak includes deals in cell therapy and oncology alongside the pain acquisition.
