Two B7-H3-directed ADCs, tambotatug pelitecan (tam-peli; YL-201) and risvutatug rezetecan (ris-rez), hit the overall survival endpoint in China-based Phase 3 trials in relapsed small-cell lung cancer. At WCLC, both studies were reported with matching hazard ratios of 0.46 versus topotecan, indicating a 54% reduction in the risk of death. The results add weight to B7-H3 ADCs at a critical moment for the class, where evidence of survival benefit is increasingly required to set new standards in a tumor space defined by limited options. The updates also highlight how China-run trials continue to shape global ADC development trajectories. Separately, Roche’s collaborator MediLink reported Phase 3 OS benefit for Tam-Peli in a China population, with additional detail: median OS of 13.3 months versus 9.4 months and OS hazard ratio 0.46 with p<0.0001.
