Eli Lilly agreed to buy 4E Therapeutics to strengthen its non-opioid pain program, adding a lead MNK inhibitor candidate (4ET1103) that entered human testing after completing a Phase 1 safety study. The Austin, Texas-based company is focused on targeting the MNK-eIF4E signaling pathway in peripheral sensory neurons to address pain without the central nervous system effects associated with opioids. Lilly framed the purchase as part of a broader strategy to find pain medicines that avoid cognitive impairment and addiction risks. 4E’s oral asset is positioned for further clinical development as Lilly continues building its pain portfolio through additional M&A. The acquisition follows Lilly’s earlier deal to acquire SiteOne in May 2025 and comes amid continued investor and clinical interest in peripheral-nerve and pathway-modulating approaches for chronic pain management.
