Eli Lilly agreed to acquire 4E Therapeutics, aiming to expand its pain pipeline with a non-opioid approach built around MNK inhibitors. The purchase targets 4E’s MNK-eIF4E signaling pathway, designed to address the shift from acute pain to chronic pain through effects in peripheral sensory neurons rather than broad central nervous system pain suppression. 4E’s lead candidate, 4ET1103, completed a Phase I safety study (NCT07586514) and is being positioned for further development under Lilly. The deal price was not disclosed. In announcing the transaction, 4E’s leadership cited Lilly’s clinical development and global commercial capacity as key to advancing the program. Lilly’s deal follows earlier pain-focused moves, including its 2025 acquisition of SiteOne, also aimed at alternatives to opioid-based treatments. The MNK inhibitor strategy is framed as a way to reduce the cognitive effects and addiction risks associated with opioid mechanisms.
