Eli Lilly has agreed to buy 4E Therapeutics, extending its push for pain treatments that avoid the brain-side effects associated with opioids. The Austin-based company’s lead candidate, 4ET1103, is an oral MNK inhibitor designed to target the MNK-eIF4E signaling pathway in peripheral sensory neurons. Lilly’s acquisition comes after earlier pain investments, including its 2025 purchase of SiteOne, and reflects a broader strategy to move beyond conventional pain mechanisms. For 4E, the deal places its early clinical-stage program into Lilly’s clinical development and global commercialization platform. The transaction is described as an expansion of Lilly’s pain portfolio through an approach intended to reduce risks such as addiction and cognitive adverse effects linked to central nervous system action.
