The iPRIME phase 2 study reported that neoadjuvant durvalumab plus tremelimumab added to standard chemotherapy failed to meet its primary efficacy endpoint in advanced high-grade serous ovarian cancer. In the dual immune checkpoint approach (Du-T-NACT), the measured CRS 3 response rate in the experimental arm did not reach the pre-specified threshold, and radiologic response rates were similar between arms. Although the trial’s secondary endpoints included progression-free survival and RECIST responses, the overall readout suggests the regimen did not deliver the chemotherapy response signal hoped for in this setting. The result is another high-scrutiny datapoint for combination checkpoint strategies moving into later testing in ovarian tumor biology.
