A preclinical study reported that IL-6-responsive engineered CAR macrophages (IL6-CD147 CAR-M) can enhance CAR-T cell antitumor activity in solid tumor models. When stimulated with IL-6, the engineered macrophages increased matrix metalloproteinase production, supporting improved tumor killing when paired with CAR T cells. In mixed-antigen mouse models, the combined therapy reduced tumor burden and prolonged survival, alongside increased immune infiltration and a shift in tumor microenvironment immune composition. The authors also highlight a key design point: IL-6 signaling is converted into a matrix-remodeling program rather than fueling extracellular IL-6. Because all data were limited to cell and animal models, the next step remains clarifying signaling mechanics and demonstrating safety with longer observation windows before clinical translation.
