A mechanistic study in PLOS Pathogens reports that the rabies virus matrix (M) protein stabilizes RGS4 to suppress neuronal ferroptosis and enable viral replication. The authors describe a regulatory axis linking lipid droplet formation, incomplete autophagy, and iron metabolic reprogramming. The work shows that inhibiting or depleting RGS4 increased ferroptosis susceptibility and restricted viral replication. Mechanistically, the study ties the pathway to changes in HERC2 and NCOA4 stability, ferritinophagy, and ultimately ferritin heavy chain loss. The findings extend ferroptosis biology into viral immune evasion and identify host signaling nodes that could be explored for antiviral development beyond classic replication-targeting strategies.
