Gilead agreed to pay $1.275 billion to acquire global rights to HCXT-2001, an oral IL-23 receptor inhibitor program from PepStar Therapeutics, which is currently in Phase 1 testing. The deal strengthens Gilead’s inflammation and immunology portfolio with a modality designed to deliver daily oral pathway inhibition. The transaction is structured as an upfront payment with additional milestones and royalties, and Gilead said the acquisition is positioned as a potentially differentiated approach to IL-23 pathway suppression for inflammatory bowel disease, including potential combination strategies. Because IL-23 is a clinically validated target in immune-mediated disease, the strategic focus now turns to whether an oral macrocyclic peptide can compete on efficacy durability, safety tolerability, and manufacturability compared with existing and late-stage IL-23 pathway drugs. The asset adds to the competitive pressure building around next-generation oral immunology pills, including other IL-23R peptide programs in development.
