Integrated DNA Technologies (IDT) advanced its CRISPR safety workflow with UNCOVERseq, a method designed to streamline off-target site nomination for subsequent confirmation. In a benchmarking study published in Nature Communications, IDT reported that UNCOVERseq generated a larger set of high-confidence sites while maintaining analytical sensitivity near 98%. UNCOVERseq is built on the GUIDE-seq protocol licensed from a Mass General spinout, SeQure Dx, and addresses a key technical limitation of GUIDE-seq—noise from non-informative double-stranded DNA byproducts enriched during sequencing. IDT said it redesigned tags to improve amplification while adding a blocker to ignore those junction byproducts. Strategically, the announcement positions IDT to offer a more end-to-end gene editing services model, including a more standardized safety assessment step that the company argues has been missing across existing off-target nomination approaches. The method is intended to perform in cell lines and under promiscuous editing conditions, aiming to better reflect native cell environments.
