SonoThera raised $125 million in an oversubscribed Series B to advance ultrasound-mediated delivery of genetic medicines into clinical development. The round was led by Vida Ventures with participation from ARK Invest, Otsuka, UCB Ventures, and major strategic investors including Johnson & Johnson Innovation and Vertex Ventures. SonoThera plans to prioritize programs in Duchenne muscular dystrophy (DMD) and autosomal dominant polycystic kidney disease (ADPKD). The company’s platform combines RIPPLE ultrasound-mediated delivery with the PORE payload engineering system, targeting delivery challenges, immune responses, and the ability to redose. The financing also sets an explicit near-term milestone: SonoThera expects to initiate its first clinical trial in DMD in 2027, making this a key test of whether non-viral, ultrasound-triggered delivery can scale clinically.
