Researchers reported that ruxolitinib, already used to dampen overactive immune signaling, may prevent irreversible autoimmune endocrine disorders in APECED. In a Nature Communications study, Pechacek, Webb, dos Santos Dias and colleagues reported findings supporting ruxolitinib’s protective effect against devastating hormone-producing organ damage. APECED is a rare inherited disorder in which immune dysregulation progressively destroys endocrine function. The work suggests that controlling immune signaling earlier—before irreversible tissue loss—could change long-term clinical outcomes. The preclinical-to-clinical relevance hinges on how rapidly intervention can be translated in human APECED, but the study adds a compelling rationale for targeting the disease’s mechanistic immune pathway with JAK inhibition.
