Genentech and DualityBio have entered a collaboration and license agreement targeting next-generation antibody-drug conjugates built on DualityBio’s DUPAC payload platform. The deal includes $45 million upfront and potential milestones that could exceed $1 billion across development and commercial outcomes. DualityBio will conduct discovery work and early-stage global clinical development through Phase Ia trials, while Genentech is set to take over for later-stage clinical development and commercialization. The partnership is framed around addressing resistance to topoisomerase inhibitor–based ADC payloads, which have shown clinical success but also face refractory biology.
