Genentech and DualityBio have entered a collaboration and license agreement targeting next-generation antibody-drug conjugates built on DualityBio’s DUPAC payload platform. The deal includes $45 million upfront and potential milestones that could exceed $1 billion across development and commercial outcomes. DualityBio will conduct discovery work and early-stage global clinical development through Phase Ia trials, while Genentech is set to take over for later-stage clinical development and commercialization. The partnership is framed around addressing resistance to topoisomerase inhibitor–based ADC payloads, which have shown clinical success but also face refractory biology. For the ADC landscape, the transaction reinforces Genentech’s continuing investment in payload innovation and diversified linker strategies to improve efficacy in heterogeneous solid tumors.
