Genentech agreed to a collaboration and license deal with Shanghai-based DualityBio built around the company’s DUPAC payload platform for antibody-drug conjugates. Roche will pay $45 million upfront, with milestone payments potentially exceeding $1 billion across programs, along with tiered royalties. DualityBio will conduct discovery work and early-stage global clinical development through Phase I, while Genentech takes over later clinical development and commercialization. The platform is designed to tackle resistance seen with earlier topoisomerase inhibitor–based ADCs by introducing payloads intended to overcome refractory disease mechanisms. For ADC investors, the transaction is a direct signal that Roche is increasing exposure to next-generation payload innovation rather than only linker and antibody engineering as the modality matures into earlier lines of therapy.
