Researchers reported preclinical evidence that engineered IL-6-responsive CAR macrophages can boost CAR-T activity against solid tumors. The construct—IL6-CD147 CAR-M—appears to increase matrix metalloproteinases after IL-6 stimulation, potentially easing immune-cell access by remodeling extracellular matrix components. In mixed-antigen mouse models, combining IL6-CD147 CAR-M with CAR-T reduced tumor burden and improved survival while altering the tumor microenvironment toward higher immune-cell infiltration and cytotoxic mediator signals. Detectable extracellular IL-6 declined, though the mechanism for that reduction remains unresolved. The findings are early and limited to cell and animal models, but they provide a concrete approach for turning a known immunosuppressive cytokine signal (IL-6) into a functional program that supports adoptive cell therapy against solid tumors.
