Researchers at Rice University unveiled CLASSIC, a combined long- and short-read sequencing approach that, together with AI/ML, enables ultra-high-throughput mapping of synthetic genetic circuit design space. The team generated millions of DNA designs, used dual sequencing to pair complete circuit sequences with short DNA barcodes, and screened circuits in human cells for function using fluorescence sorting. The advance demonstrates first-of-its-kind use of AI to design genetic circuits at scale, cutting the search time for functional DNA designs and enabling systematic exploration of multi-kilobase constructs that underpin synthetic biology applications including cell therapies and gene regulation systems.
