Researchers identified a CTCF–FADS1 molecular axis that they report predominately regulates oncogenic energy metabolism in hepatocellular carcinoma. In liver cancer models, loss of CTCF reduced proliferation, colony formation, and cell migration and invasion, alongside senescence induction. Mechanistically, chromatin profiling suggested CTCF knockout preferentially altered local promoter/enhancer binding rather than disrupting higher-order boundary organization, pointing to targeted regulatory elements as key metabolic drivers for aggressive HCC behavior.
