A preclinical study reports that intraperitoneal CAR-T cell administration improves antitumor activity in solid tumor models compared with standard intravenous delivery. Using 89Zr-oxine labeling and PET/CT tracking, researchers observed early tumor-associated radioactivity within 48 hours with intraperitoneal dosing, while end-point flow cytometry showed clearer persistence advantages and improved regional delivery. The work also suggests intraperitoneal CAR-T can influence exhaustion-associated marker patterns and trafficking dynamics. In the combination arm, pairing intraperitoneal CAR-T with anlotinib further enhanced tumor control, pointing to a possible strategy for overcoming delivery barriers that have limited CAR-T success in solid tumors. For developers, the key translational question is whether route-specific persistence and tumor infiltration can translate into durable clinical responses with manageable safety.
