First-in-human data for roginolisib, a conformation-selective PI3Kδ inhibitor, showed acceptable tolerability in advanced cancers and survival signals in metastatic uveal melanoma, reaching median overall survival up to 20.8 months in the report. The study also described immune changes consistent with relief of tumor-induced immunosuppression, aligning with the drug’s intended mechanism beyond direct tumor cytotoxicity. While phase 1 studies are not definitive efficacy trials, the combination of safety and biologically driven immune modulation will be closely watched by teams evaluating next-line PI3Kδ strategies in ocular malignancies and other PI3Kδ-sensitive settings.
