Bristol Myers Squibb disclosed that its confirmatory phase 3 Krystal-10 trial of Krazati (adagrasib) plus Erbitux (cetuximab) failed to meet primary endpoints in previously treated KRAS G12C-mutated metastatic colorectal cancer. The randomized study enrolled 461 patients and was designed to validate the earlier accelerated approval pathway. In the disclosure at ESMO GI Congress 2026, BMS said the trial missed statistical significance on both dual primary endpoints of progression-free survival and overall survival. BMS indicated the final analysis nevertheless supported clinical activity of the combination and said it is discussing next steps with regulatory authorities. The update matters for BMS’ regulatory path to converting accelerated approvals into full approvals, particularly as Krazati management continues to weigh confirmation outcomes across tumor types and line-of-therapy opportunities.
