Evommune’s phase 2b trial of EVO756 failed to beat placebo in chronic spontaneous urticaria, ending the company’s attempt to challenge Novartis in the CSU market. The oral MRGPRX2 antagonist missed its primary endpoint, with no dose producing a statistically greater effect than placebo on Urticaria Activity Score over seven days at week 12. Investigators enrolled 160 adults who were insufficiently controlled by antihistamines and randomized them to three EVO756 doses or placebo. Across doses, the UAS7 outcome did not separate from placebo, though safety and tolerability were described as acceptable. The competitive read-through is that Novartis, which already holds FDA approval for Rhapsido in CSU, is now less exposed to near-term competition from EVO756 in the same indication. Analysts also flagged that the CSU result could dampen confidence in other itch-related EVO756 programs, particularly atopic dermatitis, where read-through may be limited by disease biology. Evommune said it continues development in other planned studies, including atopic dermatitis and an upcoming phase 2b migraine prevention trial.
