Roche is discontinuing two Huntington’s disease antisense programs in collaboration with Ionis, citing missed efficacy and safety-driven trial stops. In a letter to the Huntington’s disease community, Roche said it will end the Phase 2 GENERATION HD2 study of tominersen after the asset failed to meet a key efficacy objective. Separately, Roche stopped a trial evaluating RG6496 after new data emerged from a parallel animal study. Roche said both decisions are based on the “totality of data” and described the developments as “deeply disappointing,” while also stating that the timing coincided “by chance.” For Ionis, the report compounds earlier setbacks in its portfolio after AstraZeneca-partnered Wainua (eplontersen) failed a Phase 3 ATTR-CM trial earlier the same week, with the stock reacting sharply. Roche and Ionis said they will continue collaborating on other programs.
