FibriLysin, an engineered fusion of staphylokinase and an endolysin, showed activity against fibrin-encapsulated Staphylococcus aureus abscess communities in a mouse model of fracture-related bone infection. The approach was reported to outperform vancomycin in the preclinical setting. The strategy targets a fibrin shield that can protect bacterial communities from antibiotic penetration. By combining functions associated with fibrinolysis and bacterial cell wall killing, FibriLysin aims to both breach the bio-protective matrix and directly eliminate pathogens. The work provides proof-of-concept for engineered dual-action biologics designed for infections where bacteria are embedded in fibrin-rich structures. Translational steps would still require safety, pharmacokinetics, and efficacy validation in models that better reflect human abscess microenvironments.
