Nature Biotechnology published research describing an AI framework—Algorithm-Guided Experimental design for lipid Nanoparticle Thermostabilization (AGENT)—that identifies thermostable, solid-state mRNA–lipid nanoparticle vaccine formulations in a short number of experimental cycles. The approach couples high-throughput experimentation with Bayesian optimization. The study reports that solid-state formulations retained full bioactivity after storage at 37°C for more than two months and induced antigen-specific immune responses in rodents and non-human primates that were non-inferior to freshly prepared soluble vaccines. The work also explores cold-chain-free delivery using microneedle patches. Separately in the same journal set, related reports addressed lipid nanoparticle design for larger RNA cargo and tissue targeting, reinforcing the role of formulation engineering in expanding platform reach. If the results translate clinically, thermostable solid-state mRNA could reduce distribution barriers and expand delivery options for mass vaccination and personalized therapies.
