AbbVie has signed a global licensing deal for Nxera Pharma’s gut-targeted EP4 receptor agonist, NXE0033744, betting that localized signaling can improve inflammatory bowel disease outcomes while reducing off-target exposure. Under the agreement, AbbVie is paying Nxera $100 million upfront through Nxera’s UK unit, with additional development and commercial milestones of up to $1.37 billion and tiered royalties. Nxera engineered NXE0033744 to restrict activation to the gastrointestinal tract by using an EP4 agonist designed for local effects. The company said a completed Phase 1 program showed reduced intestinal permeability in an indomethacin challenge cohort, consistent with the intended mechanism, with no concerning side effects reported during the program. The transaction uses Nxera’s NxWave drug discovery platform, which centers on designing small molecules for GPCR receptors. For AbbVie, the deal adds another immunology and gastroenterology program as the company continues to build out late-stage IBD opportunities. For the market, the economics underscore how big biopharma is valuing tissue-restricted approaches in inflammation, especially when they differentiate from systemic immunomodulation.
