A clinical immunology study found neutralizing antibodies against AAV3B in 33% to 36% of healthy adults, with intermediate titers in 19% of antibody-positive individuals. In a smaller Wilson’s disease cohort, 17.4% of 23 adults were positive with high titers. The findings suggest seroprevalence and titer distributions could influence patient eligibility and dosing strategy for liver-targeted AAV3B-based gene therapies. Investigators highlighted intermediate- and high-titer prevalence relevant to pre-screening requirements and trial enrollment. For developers, the results reinforce the need to model immunogenicity barriers early, and for clinicians the data support tighter antibody testing protocols when using liver-directed AAV platforms.
